چکیده
Thyroxine-binding globulin was isolated from human plasma by ammonium sulfate fractionation, chromatographic separations on diethylaminoethyl-Sephadex, gel chromatography, and two different electrophoretic procedures. The highly purified was homogeneous when subjected to polyacrylamide gel electrophoresis, ultracentrifugation analyses, and immunochemical determinations. The weight average molecular weight as determined by sedimentation equilibrium ultracentrifugations was 54,000 and by sedimentation diffusion data 55,000. Amino acid analyses indicated a minimum of 110 amino acid residues per molecule. By determination of the minimum in the curve for the fraction of maximum deviation from the amino acid analyses it was found that the minimum molecular weight for the polypeptide was 12,200. Carbohydrate analyses demonstrated the presence f equimolar amounts of amnnose, galactose, and glucosamine, and the carbohydrate portion constituted 7.5% of the total weight. The amino acid analyses suggested that thyroxine-binding globulin is composed of 4 subunits. Molecular weight determinations by gel chromatography in 6 M guanidine hydrochloride indicated the presence of three species of globulin with apparent molecular weights 52,000, 25,000, and 13,500, respectively. Prolonged storage in guanidine hydrochloride promoted a more than 60% yield of the monomeric species. Moreover, a half-molecule of thyroxine-binding globulin was isolated and shown to consist of two polypeptide chains of similar molecular weight...

Radioelectrophoresis for determination of thyroid hormone binding abnormalities in human serum.
This paper describes a rapid and accurate method for determining binding abilities of thyroid hormones to their corresponding serum proteins: prealbumin, albumin and thyroxine binding globulin. A tube cell agarose gel electrophoresis is used with radioactive labelled triiodothyronine or thyroxine. The distribution curve shows characteristic peaks for prealbumin, albumin and thyroxine binding gl...
متن کاملNovel Small Molecules against Two Binding Sites of Wnt2 Protein as potential Drug Candidates for Colorectal Cancer: A Structure Based Virtual Screening Approach
Wnts are the major ligands responsible for activating Wnt signaling pathway through binding to Frizzled proteins (Fzd) as the receptors. Among these ligands, Wnt2 plays the main role in the tumorigenesis of several human cancers especially colorectal cancer (CRC). Therefore, it can be considered as a potential drug target. The aim of this study was to identify potential drug candidates ...

متن کاملLack of significant binding of L-triiodothyronine by thyroxine-binding globulin in vivo as demonstrated by acute disappearance of 131-I-labeled triiodothyronine.
Since Gordon and his colleagues' (1) description of the association of thyroxine (T4) with an a-globulin [later termed "thyroxine-binding globulin" (TBG) ] in 1952, thyroid hormone transport has been the subject of intense study. The demonstration of "thyroxine-binding prealbumin" (TBPA) by Ingbar (2) in 1958 provided further understanding of the binding of T4 by the plasma proteins. It has als...
